As a child, D'Onofrio was an introvert, and mostly involved himself in his father's business of providing sound systems to community theatres. He completed his education from the Hialeah-Miami Lakes Senior High School. D'Onofrio's passion for drama led him to pursue the theatre from the University of Colorado, where he spent nearly 18 months. He also undertook a course in method acting from the Actors Studio, and the American Stanislavsky Theatre, which pocketed his introductory paid role in "This Property is Condemned" produced by Off Broadway. Juggling the theatre alongside various part-time jobs such as serving as a bouncer at the Hard Rock Cafe, and working as a bouncer to Led Zeppelin vocalist Robert Plant, constituted to some of the early struggles of D'Onofrio. He found his breakthrough with the role of Pvt. Leonard Lawrence in Stanley Kubrick's Full Metal Jacket. D'Onofrio had to send nearly four audition tapes, and gain nearly 30 kilos, to pocket the part of the mentally unstable marine in Kubrick's movie. He also featured in Adventures in Babysitting, Mystic Pizza, Household Saints, Ed Wood, Steal This Movie, Happy Accidents, "The Player," and "The Cell," just to name a few. After appearing in small roles in the television series The Equaliser, and Miami Vice, D'Onofrio found his small-screen breakthrough in 1997 with Homicide: Life on the Street, for which he was nominated for an Emmy Award. In the year 2000, Vincent pocketed the chance to play the part of Detective Robert Goren on Law & Order: Criminal intent produced by the NBC. He also received an award for the Best Actor at the Stockholm International Film Festival for the portrayal of Mike Cobb in “Thumbsucker,” which was an independent film. In the year 1998, Vincent D'Onofrio co-founded the 'River Run International Film Festival' with his father Gennaro, and sister Elizabeth in North Carolina. He was also involved in the works of the Utah Meth Cops Project in 2008, serving as the spokesperson and raising funds. D'Onofrio, like any true patriot, grabbed every chance to serve the nation, which was evident when he turned into the public figure of a gun control debate of the Citizens Crime Commission, New York. He also served as a narrator in the 2012 documentary "Heroes Behind the Badge." Since 2010, D'Onofrio has been a representative of the works Nation Law Enforcement Officers Memorial.
D'Onofrio tied the knot with the former Dutch model Carin van der Donk in 1997 and the couple is parents to Elias Gene, and Luka.
