Antonia was born in the city of London. In 2009, she graduated from the Bristol Old Vic Theatre School with a BA in acting then joined the National Youth Theatre.
In late 2011, she confirmed her departure from Misfits, stating that she had an "amazing time" on the show. She featured in a video for Coldplay's "Charlie Brown" in 2012. Thomas appeared in the 2013 film Sunshine on Leith and, she starred as Evie in the British rom-com TV series Scrotal Recall, which aired on Channel 4 and later on Netflix in 2014. In September 2017, she debuted as Doctor Claire Browne in the American television series The Good Doctor. She not only acted in the film Freedom's Name Is Mighty Sweet (2020) but also produced and co-wrote it.
