Manjula started her career with a cameo role in the 1999 Tamil film, Rajasthan, helmed by R.K. Selvamani. Prior to this, she played as Aparna in Sibi Malayil’s Malayalam romantic comedy-drama film, Summer in Bethlehem. In the initial phase of her career, she has faced negative response from Krishna Ghattamaneni’s fans, because they did not want her to enter the acting industry. She turned producer and was debuted in the 2002 National Award-winning Telugu film, Show. Manjula also starred in the film, alongside Surya and Lakshmi Ratan in the lead roles.

The film was highly acclaimed due to its brilliant screenplay and has earned two National Awards including the National Award for Best Feature Film in Telugu. In 2004, she worked in the production of S.J. Suryaah’s bilingual film, Naani. This was followed by Puri Jagannadh’s Telugu action film, Pokiri. His brother, Mahesh, starred in both films. Pokiri earned the Nandi Award for Best Popular Feature Film in 2006 and became the highest-grossing film in the Telugu film industry during that time. After a long hiatus, Manjula produced and starred in V.K. Prakash’ Telugu film, Kavya’s Diary; based in the 1992 Hollywood thriller film, The Hand that Rocks the Cradle.

In 2010, she played as Ram Charan’s sister in the Telugu romantic dramedy film, Orange. Manjula was also seen in the 2013 film, Sevakudu, alongside Krishna and Srikanth. Manjula established the Indira Productions, a production house, which she named after her mother.
