Her work in the Bengali movie Dibratrir Kabya earned her the National Film Award for Best Actress. On February 10, 1942, Madhabi Mukherjee was born in Calcutta, Bengal Presidency, British India. Madhabi and her sister Manjari were raised by their mother in Kolkata, Bengal, India, at the time. Madhabi got into theatre when she was a little girl. She appeared in several plays, including Naa and Kalarah. She made her film debut in Premendra Mitra's Dui Beaee as a young artist. In 1960, Mukherjee initially had a considerable influence on Mr. Sen's Baishey Shravan. Subarnarekha (The Golden Thread), which was made in 1962 but released in 1965, was her next significant film.

It was the third and final installment of a trilogy that examined the socioeconomic effects of partition; the other two were Meghe Dhaka Tara (The Cloud-Capped Star) (1960) and Komal Gandhar (E-Flat) (1961). She was chosen by Satyajit Ray in the early 1960s to play Arati in the 1963 movie Mahanagar (The Big City). She then played Charu in the 1964 movie Charulata (The Lonely Wife), based on the novella Nashtanir by Rabindranath Tagore (The Broken Nest, 1901). Kapurush (The Coward), Mukherjee's third and final movie with Ray, was released in 1965.
