Sudeshna's directorial debut happened in 2004 with Sudhu Tumi. Some of her directorial ventures are Cross Connection, Teen Yaari Katha, Bapi Bari Jaa, Biye Not out, Jodi Love Dile Na Prane, and Bitnoon.

Sudeshna Roy had won many awards, and her film Teen Yaari Katha was premiered at the Osian’s Cinefan Festival of Asian and Arab Cinema in 2012. It was also premiered at the Bangkok International Film Festival and Kolkata International Film Festival. Jodi Love Dile Na Prane by Sudeshna was chosen for the Indian Panorama Film Festival in Goa in 2014. It was also premiered at the Pune Film Festival of India.
