In a Punjabi Hindu household, Trilok was born as Dewan Basheshwarnath Kapoor's second son. He entered the nascent Hindi cinema business (Bollywood), like his brother Prithviraj Kapoor, in the 1930s and became one of the most financially successful performers of the time. He was born in Peshawar and later resided in Mumbai and Calcutta which was the only houses in the neighborhood, now known as Trilok Kapoor Marg, is his bungalow in Union Park, Chembur. Vicky and Vijay Kapoor were two of Kapoor's sons. Vicky Kapoor was a politician and attorney.

His debut performance was as the lead opposite Kanan Devi in the popular film Char Darvesh (1933). In the 1933 legendary hit Seeta, which Debaki Bose directed, he portrayed Luv. Prithviraj Kapoor, Gul Hamid, and Durga Khote starred in the movie as Rama, Laxman, and Sita, respectively. Trilok Kapoor rose to prominence as a leading man throughout the 1930s and 1940s, and for several years, he was one of the highest-paid movie stars. Later, from the 1970s until his passing in 1988, he began specializing in character roles. His films include Ram Teri Ganga Maili, Kachche Dhaage, Chor Chor, Darwaza, Sabot, and Purana Mandir. His son Vijay Kapoor's 1988 telefilm Akaanksha served as his final work.
