Based in Vishakapatnam, Andhra Pradesh, since 1979, he has been interested in acting from a young age. Having the experience being an actor in dramas in his locality, playing Raju in C/o Kancharapalem was his first film role. He was an employee of GMVC (Greater Vishakapatnam Municipal Corporation) but quit his job after receiving the offer to act.

Since 2018, Vepada Subba Rao has starred in several Tollywood films, including Palasa 1978 (2020), Bucchi Naidu Kandriga (2020), Colour Photo (2020), Uppena (2021), Akshara (2021), Vakeel Saab (2021), Kanabadutaledu (2021), Tuck Jagdish (2021), Shatagni-The Movie 92021), Gamanam (2021), Ammu (2022), Kerosene: A Burnt Truth (2022), HIT: The Second Case (2022), and 18 Pages (2022). For his role in C/o Kancharapalem, Subba Rao won the Jury Award for Best Actor at the Caleidoscope Film Festival, held in Boston in 2018.
