Sixteen contestants known as housemates live in a purpose-built house and are isolated from the rest of the world for a hundred days. Contestants nominate two of their fellow inmates for elimination each week, and the housemates who receive the most nominations in the eviction round would face a public vote. Eventually, one housemate would leave after being removed from the House. In the final week, five housemates were there in the house, and the viewers voted for the contestant whom they wanted to win.

Unlike the other versions of the English show, Big Brother, the Indian version of the show, uses celebrities as housemates, not people from the general public. All the contestants are locked in the same house for some given days. They compete against each other to win a cash prize by saving themselves from eliminations based on public votes. The episode broadcasted on Sunday mainly focuses on an interview of the eliminated contestant by the host. While all the rules of the series are not told to the audience, the most prominent ones are shown on the Television.

The inmates are not permitted to talk with each other in any other language except Telugu because it is a Telugu show. They always have to wear a lapel which is a microphone. They are not allowed to leave the house premises at any time unless they are evicted or decided by Big Boss. They are not allowed to sleep inside the house without the permission of Bigg Boss. They cannot discuss the nomination process or the elimination with anyone. The house for season one was lavish and set up at Lonavala.
