After the death of Jai Singh Rathod's (Anil Kapoor) wife and his attempt to put terrorist Roshan Sherchan in prison, he struggled with alcoholism three years after Day 1. In the hours before Roshan was due to be executed, Jai devised a secret plan to free him. As Haroon, Roshan's brother, threatened to release a deadly virus, Shibani Mallick and Vedant Acharya teamed up to hunt down Kush Sawant, the carrier of the virus.

Roshan was successfully freed by Jai and given to Haroon. Roshan got the full scoop on Jai and Haroon's deal to release a weaponized virus in six major cities for payment from an arms dealer. An ATU intervention ended Haroon's life, but Roshan escaped with the six vials of the virus. To avenge the death of Jai and the Indian government, he intended to release the virus in Mumbai. Meanwhile, Veer Singh Rathod and Gyan Thakkar tried to contain the epidemic at the Hotel Gateway Residency after a virus vial got discharged there. They discovered one of Roshan's former lovers through Haroon's girlfriend Maya, but Roshan had already ordered Shibani Mallick's execution, which Jai had to carry out.

The location of Roshan's daughter Vaidehi Sharma prompted Kiran Rathod to pose as her and was brought the ATU to Roshan. But when the ATU closed in, Roshan forced Siddharth Saigal to assist him by holding his kid hostage. When Siddharth offered to exchange Vaidehi for his son, the ATU stepped in, and Jai killed Roshan. Prime Minister Aditya Singhania caught the infection from Bhisham Bhowmick after it spread throughout the city. To identify the mastermind who had created a virus cure, Jai tracked down the final vials. At first, he blamed Devyani Bhowmick, but Jai later discovered that her father, Bhisham, had framed her. To individuals who were infected, treatment was made available. However, Shibani's mother Nupur shot Jai in vengeance after a video of Jai killing Shibani was published online.
