She grew up in Riyadh and Singapore due to her father's job. Her father is a Businessman, and her mother is a homemaker. She has an elder sister, Sara Jaffri, and a younger sister, Maher Jaffri, a filmmaker. She is married to Faris Rahman on 22 February 2014. She had a son named Aydin on 23 August 2019. Ainee Jafri graduated with a Bachelor's in Marketing major from McGill University in Quebec, Canada.

She worked with an advertising agency in Singapore. She started her showbiz career in Pakistan with Aag TV's teen drama Dreamers, followed by the TV Serial Zip Bus Chup Raho on Geo TV in 2012. She acted in Sila (2016) as Mariam and Aseerzadi (2013) as Mahira on Hum TV. Ainy's role in Burka Avenger was called one of the most influential fictional roles by Time Magazine.

Her first movie was a sports action film, Main Hoon Shahid Afridi as Alina, released in 2013. It was directed by Syed Ali Raza Usama and produced by Shahzad Nasib and Humayun Saeed. She then appeared in Balu Mahi (2017) as Mahi Jahangir, a film by Haissam Hussain. She did a drama serial on Hum TV, Tajdeed e Wafa (2018). Her other works include Haraam (2018) as Laila, Life Story (2016), and Shaandaar (2015) as Robin's Sister.

She won the Best Female Model of the Year Award in 2010. She was a nominee in the Lux Style Award 2018 for the Best Actress Category. She announced her comeback to TV screens after a four-year hiatus with the show Mandi (2023) as Ayesha in October 2022.
