She was born on 10th January 1947 in Lumberton, North Carolina, U.S. Afeni's birth name is Alice Faye Williams. Afeni grew up hating men in general because her father was abusive, and she thought her mother was weak when she could not stand up against him. In 1958, Afeni along, with her mother, and sister left her father and relocated to New York. She did her schooling at Bronx High School of Science. Afeni loved the street life of New York.

Afeni joined the movement and started a post office job at 19. She got impressed by Bobby Seale's speech, who was one of the founders of Black Panther in 1966. After marrying her co-worker, Lumumba Abdul Shakur, in 1968, at the age of 21, she changed her name to Afeni Shakur. Afeni means lover of people, and Shakur in Arabic means thankful to god.

She becomes the section leader of the Black Panther Party's Harlem chapter, where she had to train new members. As an activist, Afeni got jailed several times along with her group. The party used to bail her out and let her raise bail funds for the rest of the members in the jail. According to Afeni, she has mastered the art of raising funds for jailed party members.

Her first son was born on June 16, 1971. Afeni named him Lesane Parish Crooks, later renamed him as Tupac Amaru Shakur, which means Shining Serpent in Inca. Tupac was highly into music and songwriting but had a troubled lifestyle. In 1996, he was shot dead. Exactly after one year of her son's death, with the revenue from his album published posthumously, Afeni established the Tupac Amaru Shakur Foundation, which provides art programs for youth. She also started music label Amaru Entertainment to release Tupac's unreleased songs.

Afeni Shakur has traveled across the United States, making guest appearances and delivering lectures at schools and colleges. In 2007, she gave a flagship report for Black History Month at Vanderbilt University Commemoration. On 2nd May 2016, at the age of 69, she died of cardiac arrest in Sausalito, California.
