Nagamani is a supernatural serial based on the story of a shape-shifting snake. As the title suggests that the quest is for finding the Nagamani. The person or snake who gets the Nagamani becomes an influential person, who can do whatever they want. Shivani, a female shape-shifting serpent, comes to Earth from Naaglok in search of her husband Adishesha, who has taken rebirth in the form of a human. Shivani knows that Adishesha is the one who will help her to find the Nagamani. Nagmatha tells Shivani that Trishool or Trivikram is the real Adishsesha.

In the past, it was shown how Digvijay with Nandish and his followers kill Adishesha to get Nagamani. But they fail in their endeavor. Mahamaayi, Nandish’s mother is also after Nagamani. To take revenge on Digvijay, she has exchanged the sons of Nandish and Digvijay, who were born on the exact date and time. Both have the same snake mole. Trishul and Trivikram both fall in love with Shivani. But Shivani thinks and sees snake print on Trivikram and mistakes him as Adishesha. She rejects Trishool and marries Trivikram. 

Later, it was revealed that Trishool is the reincarnation of Adishesha. But he remembers nothing from the past. Will Shivani be successful in her quest? Will Digvijay and others find Nagamani before Shivani? Forms the plot. The serial is a dubbed version of a Kannada serial named Nagini 2. It premiered on Zee Sarthak on 15th February 2021
