Each of them focussed on different crime cases that occur in Baltimore City and how the prime characters resolved them.

The second installment touches upon the issues of drug abuse, and how urban people get influenced to serve as a drug peddler. In this, a man named McNulty wishes to seek revenge from his commander colleagues for sending him to work in the Marine unit. He finds dead bodies of thirteen women and successfully manages to put the blame on his ex-coworkers. Consequently, Major of the police Stan Valchek argues with Frank Sobotka, who is a leader International Brotherhood of Stevedores. Valchek orders to investigate Sobotka and assigns the work to Cedric Daniels. He succeeds and is wishing to get promoted.

Meanwhile, Sobotka gets involved with a smuggling circle so that he can raise some funds. His son and nephew began to help him. He later realizes that the death of the women are have a direct relation to him. He commences to investigate. Later, Sobotka and his son are arrested. The Greek kills Sobotka. Many people who are responsible for the peddling of the drugs are taken into custody. Avon and D'Angelo Barksdale, who were apprehended at the end of the first season, are still serving their respective punishments. D'Angelo Barksdale decides to stay away from his family because of his uncle's behavior.

D'Angelo Barksdale is killed by the Stringer. Oblivious to Stringer's deeds, Avon began to mourn D'Angelo. Stringers also sells one of Avon's costliest lands to his rival. Avon, who is unaware of Stringer's betrayal, hires a killer named Brother Mouzone. However, Stringer manages to convince Omar into killing Mouzone. Omar tries to kill Mouzone but he realizes that Stringent had been lying all this while and hence calls 911 to help Mouzone. However, he survives and shifts to another city while Stringer continues to work with the Proposition Joe. But this time, Avon is aware of Stringer's new job.
